Researchers at the Broad Institute and Massachusetts Institute of Technology (MIT) have made a significant breakthrough in mRNA therapeutics. By engineering multi-tailed mRNAs, they have developed a method that could potentially lead to more effective therapeutics. These multi-tailed mRNAs produce up to 20 times more protein and last up to three times longer in animals than their unmodified counterparts. The research, led by graduate student Hongyu Chen, introduces a chemo-enzymatic approach to add multiple end-modified polyA tails to a single mRNA molecule, enhancing its stability and translatability over time. This innovative approach could pave the way for long-lasting treatments that edit genes or replace faulty proteins.
An exciting new report in @NatureBiotech from @WangXiaoLab’s team led by @HongyuChenChem shows that mRNAs with branched poly-A tails yield much higher levels, and prolonged production, of the corresponding proteins compared with conventional single-tailed mRNA. Why can RNA only be linear or circular? It can be branched! We made multi-tailed mRNA, and we had fun! Thrilled to see our work on chemically and topologically engineered mRNA online! Using a chemo-enzymatic approach, we introduced multiple end-modified polyA tails on a single mRNA to boost its stability and translatability overtime. https://t.co/qWQ584xQvk https://t.co/n7e6Q3cTB4
